Stability of Adrenal Cortex Hormone in Ointment (¥°)

Abstract
The investigation was undertaken to determine the degradation of 1% Hydrocortisone and 0.3% prednisolone in polyethylen glycol ointment in order to observed the stability of adrenal cortex hormones.
The stability tests were conducted on tow different baches of ointments at 25¢ª, 40¢ª, 60¢ª, 70¢ª for about 4 months.
Decompositon occured at the C-17 sidechain as determined by the blue tetrazolium reaction.
I was observed that lowering of Hydrocortisone (Table ¥± , No.19) and prednisolone concentnration (Table ¥°) in ointment stored at room tempreature depends on the presence of phenol, and especially adsorptive action of zinc oxide and potatostarch.
At the same time, it was also known that diphenhydramine accelerates decomposition of prednisolone in PEG 400 solution, having unknown reaction mechanism.
